Unstablebiopolymersolutionsinevitablyleadtomis-characterizationofmacromolecularpropertiesandirreproducibleresults.Evenunderstableorquasi-stableconditions,persistentaggregatescanhamperreliablecharacterization,especiallyusinglightscatteringmethods.Variouspitfallsincharacterizingbiopolymerswereworkedthrough,includingdeterminationofsolutionstabilityzones,dissolutionkinetics,estimationoffractionofaggregatepopulations,andtherelationshipbetweenbatchandfractionationmethods.Chitosans,polyampholyticbiopolymerswithisoelectricpointaroundpH=6.0,withvaryingdegreesofcarboxymethylationwerestudied.Instabilitywasdeterminedvs.pHandionicstrengthusingahighthroughputscreeningmethod,simultaneousmultiplesamplelightscattering(SMSLS).Withstablesolutionconditionsdetermined,equilibriumbatchandmulti-detectorGPCcharacterizationofmolecularweight,intrinsicviscosity,andpolyelectrolytepropertieswasmade.Finally,afirstattemptatcontinuousonlinemonitoringofthemodificationreactionitselfwasmadeandcomparedtoFTIRanalysisofcarboxymethylationondiscretealiquots.Giventherangeofpossiblecharacterizationproblems,multipleapproacheswithindependentinstrumentsmayberequiredforreliablenaturalproductcharacterization.Onlinemonitoringofmodificationreactionsmayleadtorapidadvancesinunderstandingandpreparationofnaturalproducts.
