SARS-CoV-2 Spike Protein (S Protein) is a glycoprotein that mediates membrane fusion and viral entry. The S Protein of the SARS-CoV-2 virus binds Angiotensin-Converting Enzyme 2 (ACE-2), but with much higher affinity and faster binding kinetics . Polyclonal antibodies to the RBD of the SARS-CoV-2 S1 subunit have been shown to inhibit interaction with the ACE-2 receptor, confirming RBD as an attractive target for vaccinations or antiviral therapy . There is also promising work showing that the RBD may be used to detect presence of neutralizing antibodies present in a patient's bloodstream, consistent with developed immunity after exposure to the SARS-CoV-2 virus. Lastly, it has been demonstrated the S Protein can invade host cells through the CD147/EMMPRIN receptor and mediate membrane fusion.

Recombinant SARS-COV-2 Spike S1 Protein is expressed from HEK293 Cells with His tag at the C-terminal. It contains Gln14-Arg683.

分子量

Approximately 76.1 kDa. Due to glycosylation, the protein migrates to 115-140 kDa based on Tris-Bis PAGE result.

纯度

> 95% as determined by SDS-PAGE and HPLC.

活性

ELISA DataImmobilized SARS-COV-2 Spike S1, His Tag at 1μg/ml (100μl/Well) on the plate. Dose response curve for Human ACE2, hFc Tag with the EC50 of 19.4ng/ml determined by ELISA.

内毒素

1.0 EU per 1μg of the protein by the LAL method.

制剂

Lyophilized from 0.22μm filtered solution in PBS (pH 7.4). Normally 5% trehalose is added as protectant before lyophilization.

复溶

Centrifuge tubes before opening. Reconstituting to a concentration more than 100 μg/mL is recommended (usually we use 1 mg/mL solution for lyophilization). Dissolve the lyophilized protein in distilled water.

 

运输与保存方法

冰袋运输-20存,一年有效期

复溶后,-20 °C,在未开封状态下保存 3-6 个月复溶后,2-8°C 保存 2-7 天。

建议第一次使用时分装存,避免反复冻融
