GUIDELINESFORUSEPRODUCT:BDMatrigelhESC-qualifiedMatrix,5mlvialCATALOGNUMBER:354277BACKGROUND:Basementmembranesarecontinuoussheetsofspecializedextracellularmatrixthatarefoundatthedermal-epidermaljunction,atthebaseofalllumen-liningepitheliathroughoutthedigestive,respiratory,reproductiveandurinarytractsandthatunderlieparenchymaofendocrineandexocrineglands.BDMatrigelMatrixisasolublebasementmembraneextractoftheEngelbreth-Holm-Swarm(EHS)tumorthatgelsatroomtemperaturetoformagenuinereconstitutedbasementmembrane.1ThemajorcomponentsofMatrigelMatrixarelaminin,collagenIV,entactinandheparansulfateproteoglycan.2-3Growthfactors,collagenases,plasminogenactivatorsandotherundefinedcomponentshavealsobeenreportedinMatrigelMatrix.4-5STEMCELLS:Historically,hEScellderivationandculturingtechniquesutilizedserumand/ormouseembryonicfibroblast(MEF)feederlayers.6AnidealenvironmentforhEScellresearchconsistsofbothacellculturesurfacespecificallyqualifiedforhEScells,andaserum-free,definedmedium.BDMatrigelhESC-qualifiedMatrixandSTEMCELLTechnologies’mTeSR1(developedunderlicensefromtheWiCellResearchInstitute7),ahighqualitysurfaceandmediumcombination,createthefirstcompleteenvironmenttosupportfeeder-independentexpansionofhEScells.BDMatrigelhESC-qualifiedMatrixisanoptimizedsurfaceforyourstemcellresearch.IthasbeenqualifiedasmTeSR1-compatiblebySTEMCELLTechnologies,eliminatingtheneedfortime-consumingscreening,whileprovidingthereproducibilityandconsistencyessentialforyourhEScellresearch.Whencoupledwithavarietyofculturemedia,MatrigelMatrixhasbeenwidelyacceptedasanalternativesubstrateforthecultureofhEScells.8-9ThemTeSR1formulationisdefinedandserum-free,andhasbeendesignedtomaintainandexpandhEScellsinanundifferentiatedstatewhenusedwithMatrigelMatrixasasubstrate.Itdoesnotrequireanyfurtheradditionofgrowthfactorsorsupplements.ThemTeSR1formulationandMatrigelMatrixhavebeenshowntobeasuccessfulcombinationforculturingdifferenthEScelllinesforupto20passages.10CellsmaintainedinmTeSR1expresshighlevelsofpluripotencymarkerssuchasOct-3/4andSSEA-3,andpluripotencyofcellsmaintainedinmTeSR1hasalsobeendemonstratedbytheabilityofthesecellstodifferentiateintoallthreegermlayersintheteratomaassay.7,11SOURCE:Engelbreth-Holm-Swarm(EHS)MouseTumorFORMULATION:Dulbecco'sModifiedEagle'sMediumwith50μg/mlgentamycinMatrigelhESC-qualifiedMatrixiscompatiblewithallculturemediaSTABILITY:Stableforaminimumofthreemonthsfromdayofshipmentwhenstoredat-20°CStableforaminimumofsixmonthswhenstoredinaliquotsat-70°CRECONSTITUTIONANDUSE:ColorvariationsmayoccurinfrozenorthawedvialsofBDMatrigelhESC-qualifiedMatrix,rangingfromstrawyellowtodarkredduetotheinteractionofcarbondioxidewiththebicarbonatebufferandphenolred.Thisisnormal,doesnotaffectproductefficacy,andwilldisappearuponequilibrationwith5%CO2.ThawMatrigelonice.Oncethawed,swirlthevialtoensuretheMatrigelisevenlydispersed.Spraytopofvialwith70%ethanolandairdry.Keepproductoniceandhandleusingsteriletechnique.Dispensematerialintoappropriatealiquots,usingpre-cooledtubes,andrefreezeimmediately.Avoidmultiplefreezethaws.DONOTSTOREINFROST-FREEFREEZER.CAUTION:BDMatrigelhESC-qualifiedMatrixwillgelrapidlyat22oCto35oC.Thawovernightat4oConice.GelledMatrigelmaybere-liquifiedifplacedat4°Conicefor24-48hours.
