background:
TherehasbeenmuchuncertaintyintherelationshipbetweenPCV2andPMWSbutBolinetaldemonstratedthatKoch'spostulatescouldbefulfilledbyinoculatingcesarean-derived,colostrum-deprived(CDCD)pigswithPCV2andproducingclinicalsignsofPMWS.SimilarworkhasbeenperformedusinganinfectiousgenomiccloneofPCV2inspecificpathogenfree(SPF)pigstoproducethegrossandmicroscopiclymphoidlesionsofPMWS.AlthoughPCV2hasbeenshowntoinducePMWSlesionsinCDCDandSPFpigs,thepresenceofadditionalfactorsarerequired.

Function:

Self-assemblestoformthevirionicosahedralcapsidwithaT=1symmetry.Thisverysmallcapsid(17-22nmindiameter)allowsthevirustobeverystableintheenvironmentandresistanttosomedisinfectants,includingdetergents.EssentialfortheinitialattachmenttoheparansulfatemoitiesandchondroitinsulfateBofthehostcellsurfaceproteoglycans.Afterattachment,thevirusisinternalizedinaclathrin-,caveolae-anddynamin-independent,actinandRho-GTPase-mediatedpathwayandtrafficstothenucleus.ThecapsidproteinbindsandtransportstheviralgenomeandRepacrossthenuclearenvelope.
